Hi,
I made a letter with the so-called document class "letter"
Is that possible to define the vertical space between the closing and the signature?

The standard letter class is very poor in its capabilities. You may take a look at other suitable classes or packages at the CTAN Catalogue [1]. These solutions often offer a better access to the customization of things like you requested.
