We were finally able to get the third machine to compile after removing and reinstalling the entire Lyx/MiKTeX installation twice.
We were tangentially helped by a mention in the thread " XeLaTeX won't compile after upgrading to MiKTeX 2.9" in this forum.

I have read them. I can't make head or tail of them.
"Using the tex-text mapping is also equivalent to writing Ligatures=TeX. The use of
the latter syntax is recommended for better compatibility with LuaTEX documents."

Answering myself again for future reference:
- Some of these options (e.g. Letters) apply only to OpenType fonts which apparently Garamond, my trial example, is not.
- The key Letters=Uppercase, even if applied to an OpenType font, apparently does not put anything into Uppercase. The ...

Today the line of code that did not work in yesterday's post:
\newfontface\labelfont[Color=blue]{Garamond}
mysteriously works. Nothing else has changed, so I'm baffled as to why.
OTOH, the Letters option still has no effect, i.e.:
\newfontface\labelfont[Color=orange,Letters ...

Thanks Scott, for that mini-buffer tip. In fact, the flex-insert command worked correctly, but I was not respecting case-sensitivity. My commands were called MenuItem and Code, not menuitem and code. So now all is well and I will have to apply myself to figuring out how to use the LFUNS.
